How this score is calculated

It is built from checkable properties of the record itself: whether the article has a DOI, how many independent sources confirmed its data, whether the required fields are filled in, and how recent the latest confirmation is. Disagreement between sources and a missing identifier lower it. A value of 80 or above means the record was published without further checking; 60 to 79 means it is published at reduced confidence, with a manual check advisable. A record verified by an editor by hand is never pushed below 90 by a recalculation. This scores our data about the article, not the article itself.

Abstract

Influenza A virus remains one of the most formidable respiratory threats, with hemagglutinin (HA) playing a pivotal role in pathogenesis by mediating critical conformational rearrangements required for the fusion of viral and cellular membranes. Molecular docking-based positioning has emerged as a vital tool for evaluating the potential biological activity of compounds targeting viral proteins. In the present study, a structural and biological analysis of the molecular interactions between polyene polyketides and influenza A (H5) HA was conducted using the Protein-Ligand Interaction Profiler (PLIP) for automated contact annotation. A panel of 20 polyene compounds, including fungimycin, hamycin, amphotericin B, levorin, natamycin, and filipin, was investigated. It was demonstrated that the most active compounds exhibit a high capacity for forming multiple hydrogen bonds (4–9 per complex) and hydrophobic interactions with key amino acid residues, specifically LYS266A, ASN81B, LYS82B, and LEU297A, located within the stem region of HA. Binding energies ranged from –6.2 to –10.8 kcal/mol. The localization of polyenes at the HA1–HA2 subunit interface suggests a potential ability to block pH-dependent conformational transitions, which are critically important for the viral-cell membrane fusion process.
