Background and Aim. DNA quality is a critical precondition for reliable genomic analysis. This study evaluated the concentration and spectrophotometric purity of genomic DNA isolated from frozen whole-blood samples collected from 22 Zhabe horses in Pavlodar Region, Kazakhstan. Materials and Methods. Blood was collected into EDTA tubes, and genomic DNA was purified with the PureLink Genomic DNA Mini Kit using a silica-column procedure adapted for frozen samples. DNA concentration and purity were measured with a NanoDrop 2000 spectrophotometer. Results. DNA concentrations ranged from 95.4 to 247.2 ng/µL, with a mean of 172.8 ± 43.3 ng/µL. The mean A260/A280 ratio was 1.86 ± 0.05, and individual values ranged from 1.76 to 1.94. These findings indicate satisfactory recovery and generally low protein contamination. Conclusion. Frozen EDTA blood yielded genomic DNA of suitable concentration and spectrophotometric purity for routine molecular analyses. Before whole-genome sequencing, DNA quantity should be confirmed fluorometrically and fragment integrity should be assessed using an appropriate electrophoretic method.
